Abstract<\/strong><\/p>\n

Multiple myeloma (MM) is a haematological malignancy caused by abnormal plasma cell proliferation in the bone marrow. All MM cases are preceded by a precursor condition, monoclonal gammopathy of undetermined significance (MGUS), which does not require any treatment. Risk factors for MGUS include sex, age, pesticide exposure, and ethnicity. Prior studies have indicated an increased risk of MGUS among individuals with an autoimmune disorder. Survival of patients with MM has increased recently, mainly due to advances in treatment. However, MM patients typically suffer from comorbidities, which may influence survival. The project was divided into three parts. The aim of\u00a0 the first part was to investigate whether parental longevity affected survival among MGUS and MM patients using registry data from Sweden, which included individuals with MM and MGUS from 1988 to 2013. Using the same data, the second part \u00a0aimed to study the prevalence and impact of comorbidities on the survival of MM patients. The aim of the third part was to study whether autoimmune diseases were associated with a diagnosis of MGUS in a screened population, using data from iStopMM, a population-based screening study carried out in Iceland. Results from the first part show that host characteristics influence survival in patients with MM and MGUS, with a reduced risk of death among individuals with long-lived parents. In the second part, it was demonstrated that comorbidities are common in MM, with an increased risk of mortality correlated to growing number of comorbidities. Lastly, an analysis on autoimmune diseases and MGUS in a screened population did not show an association between the conditions. Previous studies with positive associations are not based on screened populations, and therefore subject to ascertainment bias. These results provide an important insight into the epidemiological host factors that influence survival in both MM and MGUS, and disprove the prior hypothesis of a causal relationship between autoimmune disorders and MGUS.<\/p>\n
