Abstract<\/strong><\/p>\n

Monoclonal gammopathy of undetermined significance (MGUS) is an asymptomatic precursor of multiple myeloma and related disorders which has also been associated with multiple other diseases. The aim of this thesis was to utilize alternative methods to overcome methodological issues and gain a better understanding of the clinical significance of MGUS. In the first section of the thesis, the relationship between MGUS and peripheral neuropathy and bone fractures is studied in population-based registers from Sweden. The second section of the thesis is built on the Iceland screens, treats, or prevents multiple myeloma study (iStopMM), large screening study on MGUS being performed in Iceland. The study rationale, design, and aims are discussed in detail and the assembled screened MGUS cohort used to assess the significance of MGUS in COVID-19. Results from the first section show that MGUS is associated with peripheral neuropathy that can, along with other factors, lead to bone fractures in MGUS. Results from the second section show that more than half of the Icelandic population over 40 years old has participated in iStopMM and by using the unique cohort from the study, MGUS was not shown to be associated with increased incidence or severity of COVID-19. These results are unexpected and highlight the need to study MGUS disease associations in screened cohorts. Such studies will further clarify the clinical significance of MGUS, hopefully leading to better care and outcomes for individuals with MGUS.<\/p>\n
