English abstract<\/strong><\/p>\n

The neonatal immune system is distinct from the immune system of older individuals and is prone to induce tolerance rather than pro-inflammatory responses, leaving neonates particularly vulnerable to infections and poor responders to vaccination. A large proportion of vaccine candidates currently in clinical development contain purified recombinant antigens which are often poorly immunogenic on their own. This has driven the interest in adjuvants in modern vaccines to enhance vaccine immunogenicity and efficacy. However, alum is the only adjuvant currently licensed for use in neonatal vaccines. Thus, there is an unmet need for safe and effective novel adjuvants to use in early life vaccinology, as well as defining their mechanisms of action and evaluating adjuvants\u2019 abilities to overcome limitations of the early life immune system.<\/p>\n

In this project, a comparative analysis of the potentials of the adjuvants LT-K63, mmCT, MF59\u00ae, IC31\u00ae and alum was performed to enhance induction and persistence of humoral immune responses after immunization of neonatal (7 days old) mice. Tumour necrosis factor superfamily receptors (TNF-R) BAFF-R and BCMA and their ligands BAFF and APRIL are important for the humoral immune response. It was demonstrated that in contrast to alum, the adjuvants LT-K63, mmCT, MF59\u00ae and IC31\u00ae enhanced both the induction and the persistence of humoral immune responses following neonatal immunization. As hypothesized, TNF-Rs and their ligands seem to play important roles both in induction and persistence of humoral immune responses. Upregulation of BAFF-R and BCMA and their ligands BAFF and APRIL by adjuvants associated with robust and sustained vaccine-specific responses. Thus, a light has been shed on molecular mechanisms that should be triggered by vaccine adjuvants to enhance induction and persistence of humoral immune responses following immunization in early life. These novel markers of adjuvanticity that have been defined can serve as biomarkers to be examined in discovery and assessments of potential adjuvants for use in the paediatric population.<\/p>\n
